The Agia Triada Monastery (Monastery of the Holy Trinity) is probably one of the most famous Meteora monasteries, Roger Moore was also here as James Bond on a deadly mission. The path from Kalambaka is sometimes quite steep and peppered with spectacular views. But it is also relatively well developed, so you don't really have to worry about your health, apart from the midday heat in midsummer. The tour is definitely worth it, then you deserve the view from the top (compared to the lazybones who drive up to the car park :-)). By the way, the monastery is closed on Thursdays, on other days the gates close at 5 p.m.
