The church stands on the foundations of a Gothic predecessor, which was built from fieldstone in the 13th century. In 1710, St. Mary's was given a pulpit at the behest of Frederick I, which was marked with the letters FR in a cartouche above it. It was probably the hospital chapel of the former Annenhospital, which was built here in the 15th century. This function is also supported by the fact that it was named after Saint Anne, the patron saint of the sick. The chapel was first mentioned in documents in 1575. The Trebbin Clauert tour also begins at the chapel. Like a large number of Trebbin residents, Hans Clauert is said to have died of the plague here, in the hospital outside the city gates, in 1566.
